Attachment of Cells to Basement Membrane Collagen Type IV

ثبت نشده
چکیده

Of ten different cell lines examined, three showed distinct attachment and spreading on collagen IV substrates, and neither attachment nor spreading was enhanced by adding soluble laminin or fibronectin. This reaction was not inhibited by cycloheximide or antibodies to laminin, indicating a direct attachment to collagen IV without the need of mediator proteins. Cell-binding sites were localized to the major triplehelical domain of collagen IV and required an intact triple helical conformation for activity. Fibronectin showed preferential binding to denatured collagen IV necessary to mediate cell binding to the substrate. Fibronectin binding sites of collagen IV were mapped to unfolded structures of the major triple-helical domain and show a similar specificity as fibronectinbinding sites of collagen I. The data extend previous observations on biologically potential binding sites located in the triple helix of basement membrane

Study of Basement Membrane Type IV Collagen Appearance in the Brain Choroids Plexus of Mouse Fetuses

Introduction & Objective: The brain choroids plexus (BCP) plays an important role in the cerebrospinal fluid (CSF) production, but its characterization is still incomplete. Collagen type IV, is one of the most important proteins of basement membrane (BM) and extracellular matrix (ECM) of BCP. In the present study we investigated the differential period of type IV collagen in basement membrane...

The Role of Type IV Collagen in Developing Lens in Mouse Fetuses

Objective(s) Extracellular matrix (ECM) and basement membrane (BM) play important roles in many developmental processes during development and after birth. Among the components of the BM, collagen fibers specially type IV are the most important parts. The aim of this study was to determine the time when collagen type IV appears in the BM of lens structure during mouse embryonic development. M...

Role of laminin in the attachment and metastasis of murine tumor cells.

We have studied the attachment of two murine metastatic cell lines and of a transformed, nonmetastatic sarcoma cell line to type IV (basement membrane) collagen. The metastatic cells attached preferentially to type IV collagen, whereas the nonmetastic cells attached best to type I collagen. Laminin increased both the rate and the number of metastatic cells attaching to type IV collagen, while f...

Epidermal cells adhere preferentially to type IV (basement membrane) collagen

Epidermal cells from adult guinea pig skin attach and differentiate preferentially on substrates of type IV (basement membrane) collagen, compared to those of types I--III collagen. In contrast, guinea pig dermal fibroblasts attach equally well to all four collagen substrates. Fibronectin mediates the attachment of fibroblasts but not of epidermal cells to collagen.
